Using the Molecular Devices V-max Microplate Reader.

 

 

General notes, precautions, and warnings:

 

1. The Spectramax control software on the PC has selections for different plate readers, make sure you are controlling the right instrument when you are running the software.

 

2. This instrument only operates in the visible light region. It can read at 405, 450, 490, & 650 nm wavelengths. Special filters must be installed before running at other wavelengths.

 

3. Any selection or command made on the control PC will over ride the selection made on the instrument keypad.

 

4. There are two types of extensions used with the instrument¹s files.

 

The ³XXXX.PPR² protocol files that are used to store instructions such as microplate layout, wavelengths, timed events, and other parameters that control how experiments are run and data is collected.

 

The ³XXXX.PDA² protocol files contain all the above information in addition to the data from the most recent run. Do not create files of this type without authorization from your instructor. If you need to create this type of file, select it as an option when using the ³save as² menu choice.

 

5. If you need a quick reference, pressing the MINI MANUAL key will cause the control computer to print out a one-page summary of the most important instrument commands and functions.

 

Routine Operation:

 

1. When the reader is turned on, it will automatically go through a self-test and diagnostics routine.

 

2. Start the instrument control software by clicking on the Spectramax icon.

 

3. If a large red ³X² appears in the upper left hand corner of the screen, it means that the instrument and control computer are not communicating.

 

4. Under the ASSAYS menu option, select the method that you plan to use.

 

5. Place the microplate on the carrier with the A1 well in the left, rear corner of the holder.

 

Do not load a plate if there are spilled liquids either on the bottom or sides.

 

The microplate must lie flat and sit squarely in the holder.

 

6. To start the analysis, press the RUN key.

 

7. Results will be printed automatically. Do not save data on the control computer¹s hard disk unless authorized by your instructor.
